X-ray crystallography
This technique can be used to determine the detailed, three-dimensional structure of single molecules like proteins as well as of macromolecular assemblies such as protein/protein or protein/nucleic acid complexes. Based on the scattering of X-rays through a crystal, this method requires the crystallization of the interesting molecule/s - which is often the major bottleneck of the method.
